Objectives: The objective of this study is to identify biomarkers of green bean intake and to assess the dose-response of such biomarkers.

Methods: In a randomized cross-over acute intervention study, 27 participants consumed 240g of green beans. Fasting urine samples were collected at baseline and further samples were collected at 2, 4, 6, and 24-hours post-consumption of green beans. Urinary metabolites were measured by liquid chromatography coupled to quadrupole time of flight mass spectrometry in both positive and negative ionization modes. One-way repeated measures ANOVA was performed to determine the features that changed post-consumption of green beans. Samples were further analyzed by tandem mass spectrometry for metabolite identification. Selected metabolites were assessed for a dose-response in a separate intervention study where 16 participants consumed a low (27g), medium (108g) and high (169g) portion of green beans.

Results: A total of 637 features in both ionization modes significantly changed with time (p < 0.05). Following examination of the time-response profiles, 318 features increased in intensity following green beans intake. From these, a total of 12 features displayed a dose-response relationship to increasing portion size of green beans (p < 0.05). These include multiple amino acid conjugates and a hydroxybenzoic acid, which are known to be present in green beans. Other examples include 3-hydroxysuberic acid and hydroxyabscisic acid glucuronide, which have been previously linked to other legumes.

Conclusions: This untargeted metabolomics approach identified 12 potential urinary food intake biomarkers for green beans, displaying both a time-response and dose-response. Future work will evaluate this panel’s ability to predict intake of green beans using biomarker data only.
