(P04-042-24) Leveraging AI to Improve Research Strategies Investigating the Complex Interaction Between Chardonnay Marc and Its Potential Impact on Gut Barrier Function

Objectives: The intestinal tract acts as the first line of defense against the exterior environment. The integrity of the gut lining is vital, and compromised integrity has been associated with pathological outcomes. WellVine™ Chardonnay marc is an upcycled ingredient comprised of skins, seeds and pulp. Sinrod et al. characterized WellVine™ and identified its diversity in oligosaccharides and polyphenols while noting its potential use as a prebiotic for gut health. The objective of this study was to investigate the potential of AI to improve research strategies seeking to understand the role of natural products found in WellVine™ on gut barrier function (GBF).

Methods: WellVine™'s molecular composition of 78 oligosaccharides, phenolics, and procyanidins was analyzed with the AI platform, LEAP™, to identify associations between WellVine™ and GBF. LEAP™ integrates and analyzes scientific publications and knowledge bases to create an evidence synthesis system with known and predicted associations. The LEAP™ analysis identified the compounds that are associated with GBF along with their general health impacts (e.g. positive, negative).

Results: A comprehensive profile of WellVine™’s impact on GBF was generated. Of all 78 compounds, 28 (36%) compounds have a positive impact, including 11 oligosaccharides, 2 procyanidins, and 15 phenolics. These compounds have diverse mechanisms of action, including modulation of gut tissue and mucus structure, immunity and inflammation signaling pathways, and the gut microbiome. One compound (1%) has both positive and negative impacts. The remaining 49 (63%) compounds have no association with GBF.

Conclusions: The findings argue that several WellVine™ compounds have a positive impact on GBF with diverse mechanisms of action, and most of these compounds are phenolics. WellVine™ also contains many novel and/or under-studied compounds that could contribute to improving health. Overall, WellVine™ may exhibit substantial positive impacts on GBF, warranting further in vitro and in vivo experiments. AI informed strategies may be utilized to guide future experiments.
